中华屈挠桿菌
中华屈挠桿菌由眉藻 (Calothrix) 培养池中分离得到一株宽度均一, 长度不等 (0.4-0.5 X 4.0-16.0 micrometer 以上) 的屈挠桿菌, 在0.5%胰胨液体培养基中呈长丝状,长度可超过 60 micrometer. 细胞有时聚集成团。

摘要 | 由眉藻 (Calothrix) 培养池中分离得到一株宽度均一, 长度不等 (0.4-0.5 X 4.0-16.0 micrometer 以上) 的屈挠桿菌, 在0.5%胰胨液体培养基中呈长丝状,长度可超过 60 micrometer. 细胞有时聚集成团. 兼性厌氧. 革兰氏阴性 , 无内生芽孢及其它形式的休眠细胞. 滑行运动. 能利用 多种糖类作为碳源, 但不分解琼脂,几丁质和纤维素. 可 以NO3-, NH4+, 酪蛋白水解物作为唯一氮源. 不液化明胶 . 在胰陈琼脂上形成粉红-橙色粘性菌落. 色素光谱测定 最高吸收峰值为484nm(正己烷提取), 在乙醇提取物中为 484nm, 510nm. DNA中6+C含量为34.2克分子%. 对固氮蓝 藻营养细胞有溶解能力. 对四环素以外的多种抗菌素均具 有相当强的耐受性.与已知的屈挠桿菌有明显的差异, 认 为是一新种, 命名为中华屈挠桿菌 (Flexibacter chinenses sp. nov.). 模式株FCA存于中国科学院水生生 物研究所. |
Abstract | Strain FCA was isolated from the Calothrix nursery bed in the Institute of Hydrobiology. It is a flexible rod or filament in shape with uniform width andvarious length (0.4-0.5 by 4.0-16.0 micrometer). When it is grown in the liquid medium the length is usually more than 60.0 micrometer.Sometimes the cells aggregate to form a mass. Motile by gliding, resting stages not known. Gram-negative. The colour of colonies of strain FCA is pink, with a pale iridesence, and turn to orange with ageing. But on the third day the cell mass becomes colourless.The maximum absorption peak of crude extract of pigment (in n-hexane or ethanol) is at 484nm. Facultatively anaerobic; hydrolyzes starch; many sugars are utilized and acid is produced from glucose, malatose, lactose, galactose, sucrose, xylose,anabionose, rafinose and cellobiose, but not from mannitol, fructose and glycerol. Cellulose (filter paper or carboxymethyl cellulose), chitin and agarare not attacked. Casein hydrolysate, tryptone, ammonium or nitrate alone is utilized as a sole N source. Nitrate is reduced to nitrite.Gelatin and tyrosine are not degraded. The DNA G+C content of strain FCA is 34.2 moles %. The initial isolate possesses ability to lyse vegetative cells of Anabaena strain 1105. Growth of strain FCA is inhibited by 0.01% SDS, susceptible to tetracycline 10 u/mL, resistant to 10 ug actinomycin D, 2,000 u polymyxin B or E,1,600 u penicillin G, 60 u streptomycin, 25 ug mitomycin C, 600 u erythromycin, 100 u chloramphenicol, and 200 u neomycin per mL etc. This strain belongs to the genus Flexibacter, but it differs significantly from species previously described. Therefore it is regarded as anew species, for which the name Flexibacter chinenses is proposed. Type culture FCA is deposited in the Institute of Hydrobiology, Academia Sinica, Wuhan. |
